Spike and recovery is used to determine whether analyte detection is affected by differences in the standard curve diluent and biological sample matrix. If this known substance is also present in the unknown, then the peak for that substance will be larger in the spiked sample than in the the unspiked sample. Co-injection involves adding a known substance to the unknown. You will use a process called co-injection or spiking to identify alcohols in a n unknown misture. Spiked samples may be used in method validation experiments to help identify matrix effects and determine the recovery of an analyte or the selectivity of the method.
